用于延长药物半衰期的抗体、其融合蛋白和应用的制作方法

本发明属于治疗性抗体药领域,涉及一种用于延长药物半衰期和提高药物生物利用率的抗体及其应用。
背景技术
:大多数蛋白和多肽药物的半衰期很短,一是因为蛋白质药物会被体内的蛋白酶降解而快速清除;二是由于肾小球的过滤作用使分子量小于60kda的药物在肾脏代谢中也易被清除。现有技术中,尝试了各种方法延长药物体内半衰期,包括对分子结构进行改造和缀合,然而各种分子设计都或多或少存在一些问题。以glp-1(胰高血糖样素-1)分子为例,天然的glp-1在人体血循环中可以被二肽酶ⅳ(dipeptidylpepidiaseⅳ,dppⅳ;cd26)迅速降解,其体内半衰期只有2-5分钟,不具备成药性。采用氨基酸突变、赖氨酸残基脂肪链修饰、peg化等技术可以达到每天一次用药。已经上市的此类glp-1类似物包括(1)艾塞那肽(商品名byetta)是第一个上市的glp-1类似物,皮下注射,初始计量一次5ug,一天两次;(2)利拉鲁肽(商品名victoza/saxenda)每日注射一次,每次注射0.6mg-1.8mg;(3)利司那肽(商品名lyxumia)起始剂量为10μg,每日一次,应用14天,维持剂量:在第15天开始20μg为固定维持剂量,每日一次。上述方法中,较为常用的方法是将多肽或蛋白质药物直接与化合物peg相连形成聚合物,然而这样会使药物的分子量变大,同时会增加药物的副作用。在此基础之上研制的二代glp-1类似物,采用hsa或fc蛋白融合技术,可以达到一周一次用药。例如,将多肽或蛋白质药物直接与人fc区相连融合表达的杜拉鲁肽(商品名:)是由美国lily公司研发的新型长效glp-1r激动药,由两个具有dpp-4抑制作用的glp-1类似物和人免疫球蛋白重链igg4-fc片段融合得到,其活性与内源性glp-1相似,半衰期为5天,能有效延缓肾脏的清除作用。fda于2014批准的杜拉鲁肽皮下注射液上市。每周给药一次,每次给药量0.75mg-1.5mg。然而与人fc区融合表达的药物具有引发adcc、cdc的风险。另一种方法是将多肽或蛋白质药物直接与化合物peg相连形成聚合物,然而这样会使药物的分子量变大,同时会增加药物的副作用。还有一种方法是将多肽或蛋白质药物直接与人血清白蛋白(humanserumalbumin,hsa)相连融合表达。人血清白蛋白是人体血液中的主要蛋白,该蛋白由585个氨基酸构成,是人体循环系统内的含量最多的可溶性蛋白,在血液中的浓度为34~54g/l.白蛋白是最丰富的血浆蛋白,具有高度的可溶性,非常稳定,能与人体内药物分子结合运输到体内各个部位发挥疗效。目前市场上的阿必鲁肽(albiglutide),是两个glp-1分子串联后在c端连上改造后的人血清白蛋白,建议给药量30毫克,每周一次。然而与人血清白蛋白融合表达的药物分子量较大,不具有很好的细胞穿透性。尽管目前市场上已有多种延长多肽或蛋白质药物半衰期的药物,但不是用量大就是半衰期短,仍有必要进一步开发具有延长的药物半衰期,同时成本更为低廉,患者依从性更好的药物结构。技术实现要素:本发明涉及一种延长治疗性蛋白质治疗效果特别是延长其半衰期的方法,特别是该方法中使用的具有特定结构的抗体,以及依照该方法所获得的包含治疗性蛋白质的融合蛋白。具体而言,本发明利用特定引物扩增获得的抗体cdna而构建噬菌体库,并筛选得到了与人血清白蛋白结合的一组抗人血清白蛋白抗体,该抗体在与特定治疗性蛋白质药物融合表达后可延长药物半衰期。本发明的一方面涉及一种抗体,其特征在于仅包含抗人血清白蛋白抗体的重链可变区,优选由fr1、fr2、fr3、fr4、cdr1、cdr2、cdr3组成。优选的,所述抗体的cdr区如表1的seqidno:1-28任一序列的三个cdr区的组合所示。表1如seqidno:1-28所示的抗体氨基酸序列结构优选的,所述抗体的氨基酸序列如seqidno:1-28的任一项所示,或者与seqidno:1-28的任一项具有至少70%的序列同一性,更优选的,与seqidno:1-28的任一项具有至少80%的同一性,再优选的,与seqidno:1-28的任一项具有至少90%的同一性,进一步优选的,与seqidno:1-28的任一项具有至少95%的同一性。本发明的第二方面涉及一种包含治疗性蛋白质以及上述抗体的融合蛋白。可选的,所述治疗性蛋白质选自短肽分子、抗体分子、抗体片段、细胞因子,干扰素,肿瘤坏死因子,肿瘤标志分子。优选的,所述治疗性蛋白质为glp-1或其类似物、衍生物。优选的,使用胰高血糖素样肽-1(glucagon-like-peptide1,glp-1),其是已发现的促胰岛素分泌作用最强的肠肽类激素。glp-1(7-37)的序列如seqidno:70所示:haegtftsdvssylegqaakefiawlvkgrg。glp-1的类似物或衍生物是指与glp-1具有同源性,并且具有相同生物学活性的结构类似物,包括但不限于下列药物的治疗性蛋白部分:杜拉鲁肽(dulaglutide)、利拉鲁肽(liraglutide)、索马鲁肽(semaglutide)、阿比鲁泰(albiglutide)、他司鲁泰(taspoglutide)、tirzepatide、利西拉来(lixisenatide)等。本发明的抗体片段包括但不限于,例如单链抗体(singlechainantibodyfragment,scfv)及其多价形式,fab和重链抗体。本发明的第三方面涉及包含glp-1或其类似物、衍生物,以及由抗人血清白蛋白抗体重链可变区组成的抗体的融合蛋白。作为进一步优选的实施方案,本发明的融合蛋白在抗体和治疗性蛋白质之间包含连接短肽,所述连接短肽具有下述任一种结构:(1)(gx1ggs)n其中x1为g或s,n为2-7任一值,或(2)(gggs)n,其中n=2。作为具体的实施方式,本发明融合蛋白的氨基酸序列选自seqidno:35-41。本发明的又一方面涉及一种上述抗体的筛选方法。所方法包括如下步骤:(1)以人血清白蛋白免疫羊驼(llama);(2)分离羊驼外周血pbmc,提取mrna,反转录成cdna;(3)两轮pcr扩增目的片段,构建成噬菌体库,利用elisa筛选免疫阳性克隆;(4)将阳性克隆进行诱导表达纯化,测定与人血清的亲和力;进一步的,本发明还涉及利用所述抗体制备融合蛋白的方法,其进一步包括以下步骤:(5)将阳性克隆序列与治疗性蛋白质序列在原核系统中融合表达、纯化。在上述制备方法中,使用特定的引物扩增目的抗体片段是关键的。本发明利用统一的两套引物分别进行两轮pcr扩增,实现了对目的抗体dna序列的扩增。所述引物为:引物名称引物序列kz-001caggtgaaggtcatcgartc(seqidno:29)kz-002gatgctcttgtgactcaggaatc(seqidno:30)kz-003ggaattccatatggattataaagatgatgataaacgcagagacagtgaccagagt(seqidno:31)kz-004ggaattccatatggattataaagatgatgataaacaggtcaccttgaaggagtctgg(seqidno:32)kz-005ggaattccatatggattataaagatgatgataaacaggtgcagctgcaggagtcggg(seqidno:33)kz-006ccacgattctgcggccgcttactgaggagacagtgacctgggtcc(seqidno:34)其中,第一轮扩增使用引物kz-001和kz-002,第二轮扩增使用引物kz-003、kz-004、kz-005和kz-006。本发明的融合蛋白具有治疗活性,根据所包含的治疗性蛋白的生物活性,所述融合蛋白表达后保留甚至提高了原有生物活性,其可用于原有治疗性蛋白所覆盖的任何疾病以及治疗领域。优选的,本发明结合了glp-1或其类似物、衍生物的融合蛋白可以用于治疗糖尿病,更优选为非胰岛素依赖性糖尿病。所述融合蛋白还可以具有以下治疗活性:减少体重:glp-1主要通过作用于大脑与胃肠道起到减肥效果,一方面其可降低大脑的进食欲望并使人体的能量消耗加强,另一方面可减缓胃排空增加饱胀感,减少患者进食,从而达到降体重的效果。心血管受益与肾脏保护:glp-1可通过降血压、加强心肌收缩与肾脏的钠排泄等作用机制从而达到轻度降压、保护肾脏的作用,可降低多重心血管危险因素,有效减少心血管事件的发生(参考文献:diabetescare.1998;21:1925-1931;diabetessectrum,2004;17:183-190;diabetessectrum.2004;17:183-190)。技术效果:本发明实现了将抗体与蛋白质药物融合表达,在保留甚至进一步提高了蛋白质药物药效的前提下,延长了药物的半衰期,提高了生物利用率,同时相比其他蛋白质药物改构方式而言,工艺简单,成本低廉。具体而言,本发明在以下方面获得了有益效果。1.产品结构与性质方面与fc区或完整hsa融合蛋白形式相比,本发明的抗体融合蛋白是新一代glp-1类似物。将glp-1与本发明的抗体融合,具有分子量小,易穿过屏障的优点,避免了fc区融合蛋白具有的引发adcc、cdc的风险。更为重要的是,依照本发明所获得的融合蛋白相比其他改构形式具有更长的半衰期,前期数据表明,半衰期能达到两周甚至更长。如

发明内容,但本发明的范围不限于此。如无具体说明,以下实施例中使用的试剂和仪器都是本领域常规试剂和仪器,可以通过商购途径获得;所使用的实验方法是常规实验方法,本领域技术人员根据现有技术完全可以知道如何重复所述实验并获得相应结果。实施例1:免疫羊驼及其效价测定将人血清白蛋白多点皮下注射进羊驼体内,经多次免疫后取少量血清测效价(酶联免疫法,靶抗原为人hsa蛋白,一抗为抗his-hrp,tmb显色,450nm检测o.d值)。当效价达到血清稀释32万倍后酶联免疫结果在1.0以上即可。检测时将血清用ph7.4pbs缓冲液稀释,其中血清被稀释倍数为100倍、1000倍、10000倍、40000倍、8万倍、16万倍、32万倍、64万倍。表2不同血清稀释倍数下的效价测定结果血清被稀释倍数1:1001:1k1:10k1:40k1:80k1:160k1:320k1:640k空白孔elisa结果3.2623.3163.4583.4263.5083.2963.171.020.057实施例2:构建噬菌体库及筛选1.cdna的获取对实施例1获得的血清满足效价要求的羊驼进行大量采血,分离pbmc,用15×106个pbmc细胞提取总rna,提取总rna的方法为trzol手提操作法。使用thermo公司反转录试剂盒将总rna反转录成cdna,将得到的cdna混匀分装备用。2抗体目的片段的获取利用下列2组pcr引物进行两步pcr扩增,获得目的片段。引物名称引物序列kz-001caggtgaaggtcatcgartckz-002gatgctcttgtgactcaggaatckz-003ggaattccatatggattataaagatgatgataaacgcagagacagtgaccagagtkz-004ggaattccatatggattataaagatgatgataaacaggtcaccttgaaggagtctggkz-005ggaattccatatggattataaagatgatgataaacaggtgcagctgcaggagtcgggkz-006ccacgattctgcggccgcttactgaggagacagtgacctgggtcc反应试剂如下:反应程序如下:以kz-001和kz-002为引物进行第一轮pcr,用takara公司高保真pcr聚合酶进行扩增,扩增程序如下:94℃预变性3分钟,(94℃变性30秒,53℃退火30秒,72℃延伸40秒)循环18轮,72℃末端延伸10分钟,4℃降温1分钟。回收750bp条带。以kz-003、kz-004、kz-005和kz-006为引物进行第二轮pcr,用takara公司高保真pcr聚合酶进行扩增,扩增程序如下:94℃预变性3分钟,(94℃变性30秒,53℃退火30秒,72℃延伸30秒)循环20轮,72℃末端延伸10分钟,4℃降温1分钟。回收450bp条带。.3抗hsa抗体库的构建与筛选经过两轮pcr扩增得到大量目的片段后进行sfii/noti双酶切,使用商业化t4dna连接酶将酶切完成的目的片段连入噬菌体载体中,之后电击转入tg1感受态。建库库容为1.07×108,随机挑取20个克隆进行测序,测序结果分析显示,插入的片段皆为重链可变区片段,阳性率100%,包装成噬菌体库,滴度为:2.15×1012pfu/ml。采用固相筛选方法筛选噬菌体库。筛选流程是将靶分子包被在96孔表面用,洗去未吸附的靶分子,然后封闭,之后用扣除背景的噬菌体抗体库加入到孔中结合。洗去未结合的噬菌体,再用0.2m甘氨酸-盐酸洗脱得到亲和噬菌体。每轮通过降低包被靶分子浓度和加大洗涤力度(本发明是减小靶分子浓度增加洗液中吐温20浓度)得到高亲和力噬菌体。每经一轮淘选扩增试验,就会使能与靶分子结合的抗体在噬菌体库中得到富集。经过3轮筛选后富集达到1000倍以上,进行挑取单克隆验证。3轮筛选,筛选结果如下:表3富集筛选结果筛选轮数投入量产出富集空白扩增后滴度12.15×10113.58×1076.01×1048.5×1063.06×101223.06×10111.24×1072.47×1041.4×1062.33×101332.33×10111.64×1081.42×1032×1051.62×1013从第3轮筛选产物中挑取80个单克隆进行elisa鉴定,elisa鉴定方法如下:用ph9.6包被液稀释靶蛋白hsa,37℃静置1小时进行包被,pbs洗3次。加入封闭液进行封闭37℃静置1小时,甩出多余的封闭液pbs洗3次。取扩增产物用1%的m-pbs(脱脂奶粉-pbs)10倍稀释混匀50μl/孔37℃静置1小时。一抗:用1%m-pbs按1:1000稀释兔抗m1350μl/孔37℃静置1.0h。二抗:用1%的m-pbs按1:3000稀释hrp-羊抗兔50μl/孔37℃静置1.0h。显色:取0.2mna2hpo4 0.1m柠檬酸各4.5ml加入少量opd,60μlh2o2混匀,50μl/孔。2m硫酸终止50μl/孔。490nm检测。phageelisa最少要重复一次。阳性克隆保存:取0.5mlphage (2)分离羊驼外周血pbmc,提取mrna,反转录成cdna;

(3)两轮pcr扩增目的片段,构建成噬菌体库,利用elisa筛选免疫阳性克隆;

(4)将阳性克隆进行诱导表达纯化,测定与人血清的亲和力;

(5)将阳性克隆序列与治疗性蛋白质序列在原核系统中融合表达、纯化,

其中,两轮pcr所用引物分别为seqidno:29-seqidno:30以及seqidno:31-seqidno:34。

10.权利要求6-8的融合蛋白在制备治疗性药物中的用途,优选的,所述治疗性药物用于治疗非胰岛素依赖性糖尿病,或者所述治疗性药物为减肥药或心血管疾病治疗药。
